I'm going to Japan for 14 days in early June, arriving in Osaka and ending my trip in Hokkaido. Most likely spending 3 or 4 days in Osaka and Kyoto, 4 days in Tokyo, and at least 5 days in Hokkaido. Any recommendations for first time traveler to Japan?

You should definitely not miss typical sightseeing places such as Kiyomizu Temple in Kyoto and Sensoji Temple in Tokyo. 
Here is my opinion what to not miss:

- Kyoto
Beautiful temples and shrines, historic architectures, Kyoto style Kaiseki cuisine

- Osaka
Lively atmosphere, Osaka food (Okonomiyaki and Takoyaki), Osaka Castle

- Tokyo
Izakaya (Bar hopping), don't miss numerous and various places to eat and drink, shopping

- Hokkaido
Great nature and seafood and fresh vegetables, do something to enjoy Hokkaido's nature. Visit Biei to see the beautiful flower field. If you have a chance, hiking in Shiretoko's is extremely beautiful.
